Mitochondrial methylation in health and disease

Abstract
Methylation is of central importance for cellular function, ranging from epigenetic regulation of gene expression to the maturation of neurotransmitters & metabolites. Mitochondria form a central hub in cellular metabolism but surprisingly little is known about the role of protein methylation inside mitochondria. By generating patient-specific Drosophila melanogaster models, with defects in the mitochondrial S-adenosylmethionine (SAM) carrier, SLC25A26, we were able to identify two distinct cellular responses to intra-mitochondrial SAM (mitoSAM) deficiency. Severe depletion of mitoSAM leads to an acute shortage of small molecules and metabolites, while a mild mitoSAM deficiency resulted in a slowly progressive OXPHOS deficiency and shortened life span. By applying methyl-SILAF (stable isotope labelling of amino acids in flies) in combination with mass spectrometry we were further able to map the mitochondrial methyl-proteome. Correlating the proteomic changes observed in the mitoSAM-deficient fly lines to the individual methylation sites allows us to dissect important functional roles for the mitochondrial methylproteome.
