Abstract

Laser scanning multi-photon microscopy (MPM) is a powerful tool for non-invasive three-dimensional and of biological tissue. MPM operates in the near-infrared optical window, where the absorption in tissue is low. The localized region enables confocality without the use of a pinhole while providing less photobleaching and photodamage.

A limitation associated with multi-photon microscopy is the generation of out-of-focus fluorescence when imaging deeply in highly scattering tissue, which limits imaging depth. Annular laser beams have interesting properties, and could provide a way to reduce out-of-focus fluorescence. In this thesis, mathematical models have been implemented to investigate the potential of this technique. It could be concluded that illumination with annular beams provides a lower out-of-focus fluorescence than with Gaussian beams, while retaining the shape of the focal volume. In addition, an experimental MPM was set up and initial experiments were performed on a simple tissue phantom. This first proof-of-principle experiment demonstrates a reduction of out-of-focus fluorescence of 5%. Future work will be done to further reduce out-of-focus fluorescence by improving the set-up.

Multi-photon induced luminescence in gold nanoparticles was explored for use in multi-photon microscopy. Gold nanoparticles have previously been explored as contrast agents, and it is therefore important to understand the process by which they exhibit luminescence. The study was carried out using gold nano-particles deposited in a gradient, which were imaged using a commercial MPM setup with settings applicable to life sciences. It was concluded that 10 nm gold nano-spheres need to be clustered to be visible using a multi-photon microscope, which is important to take into account when working with this type of materials.

Biomedical photonics have identified targets of haptenation in human skin


Our work has been highlighted in Current Optinion in Immunology by Stefan F. Martin.
The first in vivo targets of haptenation in human skin were identified in an elegant study. Caged fluorescent haptens, thiol reactive bromobimanes, were used to detect target proteins in full thickness human skin 20 h after application using two photon fluorescence microscopy [Simonsson et al, 2011].
